The metabolism of the intervertebral disc in rats, using a self-destructing disc model, is described. The disc is incubated in situ in Krebs-Henseleit buffer and analyzed at intervals of 5 min or 2 h. The results obtained are compared with predictions made by the method of Lieber et al., using the spontaneous turnover of the nucleus pulposus. The model simulates the metabolism of collagen and proteoglycan in the disc in vitro, and so has been adapted for the model of the disc. At 10 min, there is a low turnover of the disc (approximately 10% per hour), with a constant concentration of interstitial fluid, while the concentration of hydrated collagen is constant. The rapidity of this turnover corresponds with the experimental findings of Seki and Nashimoto. At 2 h, there is a higher turnover of the disc (50% per hour), with a slower turnover of collagen and interstitial fluid. The turnover of both proteoglycan and collagen is constant at 24 h.
